The Town Two Clinic in Khayelitsha is expected to reopen next Monday after it was temporarily closed due to safety concerns.
The City of Cape Town said the closure followed serious threats against security staff.
This included an incident where two guards were held for ransom by extortionists who demanded money from a private security company.
The matter was reported to the South African Police Service (SAPS).
The City of Cape Town said while the clinic was closed, alternative arrangements were made to ensure residents continued receiving care.
A mobile clinic has been operating in Mandela Park, providing chronic medication, immunisations, and reproductive health services.
Some patients were referred to nearby clinics in time of need.
The city said the safety of patients and staff remains its top priority. The municipality is urging anyone with information to contact the police.
