The 35-year-old and his co-accused, 25-year-old Luxolo Mbhokwe, are accused of killing Sergeant Simphiwe Kwepile in June 2023.
Mbokwe, who is in custody, was arrested in September 2023, and his case is now before the Khayelitsha Regional Court.
Gugushe emerged from the holding cells to a packed gallery filled with members of his family.
He was wearing a black jacket, blue jeans and T-shirt, and slides on his feet.
His lawyer told the court that his client would be making a formal bail application, which the prosecution indicated it would oppose.
The State said that Gugushe faced a serious charge of murder and believes that should he be released on bail, he might intimidate witnesses.
Gugushe is set to be back at the Khayelitsha Magistrates Court on 18 February to formally apply for bail.
