The attack comes three weeks after 18 relatives were gunned down at their family home in Lusikisiki.
Police spokesperson Siphokazi Mawisa said a manhunt is underway for the perpetrators.
"The Eastern Cape Provincial Commissioner Lieutenant General Nomthetheleli Mene has mobilised all available resources to find the perpetrators behind the killing of five people at Bityi on Friday night, 18 October at about 20:30 at Ncenjana location."
DECLINE IN MORAL VALUES
The ANC in the province said the mass shooting reflects a decline in moral values within communities.
The party said the attack highlights the reality that criminals have no regard for human life or the law.
It has called on residents to assist the police with any information regarding the attack.
"The provincial government condemns in the strongest possible terms the shooting and killing of five people in Bityi. The MEC (Xolile Nqatha) is making a clarion call to all the people to work with police and report these brazen criminals so that they can be arrested and account for their actions," said
